Two-color Flow Cytometric Analysis Lymphocyte Subesets in Peripheral Blood of Normal Pregnant Women



Abstract
To define alteration of peripheral blood lymphocyte subsets during pregnancy, the author analyzed helper T cells, suppressor inducer T cells, suppressor T cells,cytotoxic T cells, activated T cells, B cells and NK cells in 35 cases of pregnant
women
divided into 1st 2nd and 3rd trimester, and in 15 cases of healthy non-pregnant women as control. The lymphocyte subsets were analyzed by flow cytometry with two-color immunofluorescent stain using monoclonal antibodies. The percentage of
CD3+cells
was
significantly decreased in the 1st and 2nd trimester as compared with non-pregnant women. The percentage of CD4+cells was significantly decreased through pregnancy, especially in the 1st and 2nd trimester, and the absolute count was significantly
decreased through pregnancy. The absolute number of CD8+cells was significantly decreased in the 2nd trimester. The absolute number of helper T cells(leu8a+Leu8-) was significantly decreased through pregnancy. The percentage of suppressor inducer
T
cells (Leu3a+Leu8+) was significantly decreased through pregnancy, especially in the 2nd trimester, and the absolute number was significantly decreased through pregnancy, especially in the 1st and 2nd trimesters. From these results, it is
suggested
that
a numerical imbalance of T cells subsets, especially of both helper T cells(Leu3a+Leu8-) and suppressor inducer T cells (Leu+3a+Leu8+) has a role in the immunodeficiency of pregnancy.
